- Specificity: The enzyme releases O-linked β-N-acetylglucosamine (GlcNAc) from serine and threonine residues of glycoproteins and glycopeptides. No detectable activities of proteases, exo-β-N-acetylglucosaminidase, or other glycosidases are observed.
- Content: Purified lyophilized powder
- Reconstitution: Dissolve the lyophilized product in 100 μL of ultrapure water to make a 10,000 units/mL in buffer.
- Buffer: 50 mM Tris, 100 mM NaCl, pH 7.5
- Storage: -20 °C as powder. Once reconstituted, 4 °C for five days or -20 °C for up to three months. Avoid freeze-thaw cycles.
- Unit Definition: One unit is defined as the amount of O-GlcNAcase required to catalyze the release of 1 nmole p-nitrophenol (pNP) from p-nitrophenyl-N-acetyl-β-D-glucosaminide (pNP-GlcNAc) per min at 37°C in 100 μL 1X Reaction Buffer (50 mM Tris, 100 mM NaCl, pH 7.5).
