PNGase F

PNGase F 能有效地從醣蛋白中切除幾乎所有 N-link 寡醣。PNGase F 屬於酰胺酶 (amidase) 的一種,能特異性地在 GlcNAc 與 asparagine 之間進行切割,並完整保留 N-link 寡醣的主要核心結構,使反應後的產物適合更進一步的分析。唯當醣基中存在 α1,3-linked core fucose 時,會大幅干擾 PNGase F 的活性。

  • Specificity: The PNGase F enzyme catalyzes the cleavage of asparagine-linked oligosaccharides, except those containing an α1,3-linked core fucose, from glycoproteins and glycopeptides. No detectable activities of proteases or other glycosidases are observed.
  • Content: Purified lyophilized powder
  • Reconstitution: Dissolve the lyophilized powder in 100 μL ultrapure water to make a 40,000 units/mL solution in the Buffer.
  • Buffer: 20 mM Tris, 50 mM NaCl, 1 mM EDTA, pH 7.5
  • Storage: -20 °C as powder. Once reconstituted, 4 °C for ten days  or -20 °C for up to six months. Avoid freeze-thaw cycles.
  • Unit Definition: One unit is defined as the amount of PNGase F required to deglycosylate 1 nanomole (15 μg) of denatured RNase B in 1 h at 37°C in 25 μL 1X Reaction Buffer (20 mM Tris, 50 mM NaCl, 1 mM EDTA, pH 7.5).
